Human Domain Antibodies with Tailored Serum Half-Life.
Domain Antibodies are engineered to allow precise control over biophysical properties and in vivo half-life to create the optimal safety and efficacy product profile. The pharmacokinetic profile of Domain Antibody products can be customised to achieve the desired therapeutic effect by a number of validated methods. Domantis has successfully conducted in vivo PK and efficacy studies on anti-serum albumin dAb™ conjugates and PEGylated dAbs.
Human Domain Antibodies for Diverse Therapeutic Targets.
In addition to the targets that are suited to conventional antibodies, dAbs can be used to address other types of targets. For example, many cell surface receptors are not amenable to antibody therapeutics because conventional IgGs bind dimerically, creating the potential for cross-linking and activation of the receptors. Domain Antibodies are especially suited to inhibit such receptor targets since they bind monomerically and do not cause receptor cross-linking even at high concentration. Furthermore, the compact binding sites of Domain Antibodies allow binding to standard antibody targets as well as to less accessible targets, such as receptor binding clefts or enzyme active sites. In addition, the small size of dAbs should allow for better tumour targeting and tissue penetration, as well as pulmonary delivery to address lung diseases.
Injectable and Non-injectable Human Domain Antibodies.
Domain Antibodies are uniquely suited to both delivery by injection and delivery by non-injectable routes such as pulmonary or oral administration. These routes are amenable because, unlike other antibody molecules, dAbs are highly soluble, stable when freeze-dried or mixed with a range of excipients, resistant to proteolysis in the colon, and suitable for encapsulation. The small size of dAbs should allow sufficient doses to be delivered by the pulmonary route to topically treat diseases of the lung (such as asthma) and by oral delivery to treat disorders of the gastro-intestinal tract (such as inflammatory bowel disease). These delivery options should translate into better patient compliance, increased potency and fewer side effects compared with conventional antibodies.
